anti-parasitic and anti-anaerobic bacterial
  • Spiramycin is a macrolide bacteriostatic antibiotic with an antimicorbial spectrum similar to that of erythromycin and clindamycin.
  • Metronidazole a 5-nitroimidazle derivative is active against various anaerobic bacteria and protozoa .
  • The complementary combination of  Spiramycin and Metronidazole shows synergism in inhibiting certain sensitive bacteria involved in buccodental infections. 
  • Spiramycin is incompletely absorbed from gastro-intestinal tract and is widely distributed in  the tissues. It has a half- life of about 8 hours.
  • Spiramycin is metabolised in the liver to active metabolites; substantial amount are excreted in the bile and about 10% in the urine. It is distributed in to breast milk.
  • Metronidazole is readily absorbed after oral administration and bioavailability approaches 100%.
  • Metronidazole has a large volume of distribution in the body. It  crosses the placenta.
  • Metronidazole is metabolized in the liver and It’s elimination half life is about 8 hours.
  • The majority of  dose of Metronidazole is excreted in the urine mainly as metabolites; a small amount appears in the faeces.

Each film coated tablet of Dentacin® forte  contains: 

  • Spiramycin           150000 IU
  • Metronidazole        250 mg 

Each film coated tablet of Dentacin® contains:

  • Spiramycin          750000 IU
  • Metronidazole       125 mg 

 Dentacin® is highly effective in the treatment of acute, chronic and relapsing stomatologic infections such as:

  • Buccodental infections due to aerobic and anaerobic germs sensitive to Spiramycin and Metronidazole.
  • Dental infections as in dental abscesses, stomatitis, aphthous ulcer, perimaxillar cellulites, pericoronitis, periodonitis and eriodontal infection. 
  • Parotiditis and submaxillaritis.
  • Pre and post-operative prophylaxis in odontostomatologic surgery.  
  • Tonsilitis and gingivitis .
  • Dentacin® can be  efficacious in  the  treatment of  those patients with AIDS and diarrhea in which cryptospordium is identified.  
  • Hypersensitivity to Spiramycin and/or Metronidazole.
  • Concomitant use of Ergotamine or other vasoconstrictor derivatives of ergot.
  • Concomitant use of Disulfiram.
  • Spiramycin Inhibit the absorption of Carbidopa accompanied by decrease in plasma levels of levodopa, hence, the dose adaptation and clinical monitoring of levodopa is recommended.
  • Drugs that may affect Metronidazole include Barbiturates and Cimetidine.
  • Drugs that may affected by Metronidazole include Anticoagulants, Disulfiram, Hydantoins and Lithium.
  • Spiramycin may increase serum Theophylline concentration.
  • Spiramycin can increase the effects of oral Anticoagulants and Digoxin.
  • Dentacin® should not be used in the first trimester of pregnancy and lactation.
  • Lactating women those must use Dentacin® should stop lactation during treatment period. 

Dentacin® forte tablets are well tolerated.

The most common side effects are:  

  • Mild gastrointestinal disturbances as nausea, vomiting, diarrhea, unpleasant taste and darkening of urine may occur.
  • Rare case of rash, vertigo, ataxia and leukopoenia.

Dentacin® is recommended to be used during meals.
Children:

  • 6-10 years : 1 tablet of Dentacin® 2 times daily.
  • 10-15 years : 1 tablet of of Dentacin® 3 times daily.

Adults:

  • 2 tablets of Dentacin® 2-3 times daily.
  • Or one tablet of Dentacin® forte 2-3 times daily.
  • The treatment should be discontinued in case of ataxia and vertigo.
  • Alcoholic drinks should be avoided during treatment.
  • Dentacin® should be used with great care in patients with blood dyscrasias or with active diseases of the central nervous system. 
  • Dentacin® should be used cautiously in patients with hepatic dysfunction.
  • The dosage of oral–anticoagulants( e.g. warfarin) should be adapted when associated with Dentacin®.
